Maybe I 'm to stupid, but is there a way to login in web_ftp (or isp_file) as a simple mailuser like https://domain.tld:81/ftpuser/ or better https://domain.tld/ftpusers/user.name?

No, you always have to be logged in in ISPconfig and go to the Web-FTP module.
